Matthews opened the scoring on eight minutes when Marta Boabonay won a free-kick, which was swung in by Makeba Black and she converted at the back post with a clever first-time finish.
She then doubled her tally for the day on 14 minutes when the Bees counter attacked at speed. A long ball over the top found Boabonay with space on the channel and she crossed to Matthews who, again, tucked home at the back post.
Brentford were not home and dry yet though, as the home side scored on 21 and 32 minutes to make the game level at 2-2 at half-time.
On 52 minutes, Matthews had her hat-trick when the Holmesdale keeper misjudged a cross and was punished by the forward firing in off the post for her third.
With just under 20 minutes to play, Matthews rounded off an exceptional individual display to wrap up the three points for the Bees, when Freya Morgan slipped her in and she slotted the ball past the keeper to make it 4-2.
Tommy Ryan-Maynard's side have won all their opening fixtures to start the season in emphatic fashion, scoring 14 times across four games.
