Marquinhos will miss Brazil's friendly against France on Thursday because of hip pain. According to Globo.com, the Paris Saint-Germain centre-back reported discomfort in the area.
He skipped ball work in Tuesday's session and had already been removed from part of Monday's training.
Carlo Ancelotti said Marquinhos has an issue but could feature against Croatia. He added that this is a demanding stage of the season and that minor problems are common.
In the same interview, Ancelotti confirmed three of his back four, full backs Wesley and Douglas Santos, and Léo Pereira, who will make his Brazil debut. He is still weighing up the other centre-back.
The alternatives in central defence are Bremer, Ibañez and Danilo.
Brazil have already lost Gabriel Magalhães from this squad. The defender was withdrawn with knee pain.
Kick-off is at 17:00 Brasília time in Boston, then 21:00 in Orlando against Croatia.
