Fabian Schär's suspension has opened the door for the Dutch defender to make his Premier League comeback after recovering from surgery on his anterior cruciate ligament (ACL) nine months ago.
Botman has played for the club's under-21s in recent weeks and will now partner Dan Burn in central defence at the Tottenham Hotspur Stadium.
The other change to Monday's win at Manchester United sees Tino Livramento come in for Kieran Trippier, who came off in the second half at Old Trafford. Trippier is, however, among the substitutes.
Martin Dúbravka is eyeing a fifth straight clean sheet in the Premier League while Alexander Isak - nominated for the league's EA Sports Player of the Month award alongside team-mate Jacob Murphy - has eight goals in his last seven games.
He also scored the winner when the two sides met at St. James' Park in September.
