Alexander Isak scored what proved to be the decisive goal as he scored for the seventh league game in a row to complete a double over Spurs and keep Eddie Howe's side in fifth place.
There had never been a 0-0 draw between the two sides in the Premier League - so perhaps it was no surprise that there were two goals inside the opening six minutes.
In recent weeks Newcastle have made a habit of going ahead early in games but this time it was Tottenham who struck first, as Dominic Solanke stooped to head home from Pedro Porro's excellent cross.
But within two minutes, the Magpies were level. Joelinton won possession, with the ball appearing to strike his arm in what was a natural position by his sides, and he fed Bruno Guimarães, who played in Anthony Gordon to drill home with an inch-perfect finish.
In the 11th minute, Sven Botman - making his first appearance since March 2024 following knee surgery - made a vital block from Dominic Solanke before Martin Dúbravka comfortably saved Pape Matar Sarr's follow-up.
Then ten minutes later Botman broke up a Spurs attack allowing Guimarães to find Isak, who threaded the ball through to Gordon from a similar position to his goal, but this time debutant goalkeeper Brandon Austin saved.
Pedro Porro fired over from range in the 28th minute but Newcastle were on top and from a low Jacob Murphy cross from the right, Isak - at full stretch - couldn't quite force home from close range.
However, with half time approaching, the Sweden striker put United ahead from a similar position. Murphy again centred and although Radu Drǎgusin got a touch, so did Isak for his ninth goal in eight matches.
The home side started the second half strongly, just as they had the first, and Dúbravka produced a good save from Sarr before Brennan Johnson hit the post from the loose ball.
But United could have killed the game off when Sandro Tonali waltzed into the box and saw his shot cannon off Archie Gray, only for Gordon to blaze over from close range.
Substitute James Maddison curled an effort inches wide in the 81st minute then Sergio Reguilón flashed a shot across goal as Spurs put Newcastle under pressure in the closing stages.
Two Solanke headers - one which dropped onto the roof of the net, the other saved by Dúbravka - were the closest they came to a late leveller either side of Harvey Barnes almost adding a third for the visitors as Howe's men saw out 12 minutes of added time to claim another three points.
