Habeebat Gbajabiamila had the first clear sight at goal, latching on to a Maddie Phillips through ball but placing her strike too close to Dannielle Parks.
Chelsie Berry and Maï Louvrier both had sighters from distance but couldn't beat the Benfica keeper.
Esme Alaneme found some defence splitting passes from right-back but the Bees' final product was not as sharp as it had been so far this season.
Then, on the brink of half-time, Berry scored her fourth of the campaign to make it 1-0. Liberty Morrice-Bartlett fed her the ball on the edge of the box and after an intricate turn, Kasha Petit returned the ball to Berry who rifled home past the frozen Parks.
Evie Thorne Ford had little to do in the Brentford net but was controlled with the ball at her feet in just her second start since joining from Millwall Lionesses.
On 53 minutes, the Bees doubled their lead when Freya Worsley did well to get to the ball before Parks and the referee pointed to the spot. Brentford captain Phillips stepped up and calmly tucked the ball into the bottom-right corner from 12 yards to make it 2-0.
Phillips then went close again from a long-range free-kick and Berry had a chance that she put just wide seconds later.
With 20 minutes to play, second-half substitute Khyri McKenzie made it 3-0 with her third of the campaign, lifting the ball over Parks after clever work from Renai McCrea to keep the chance alive.
Williams' side were denied a fourth on 78 minutes when Asees Sangha's fierce strike was saved excellently by Parks diving low to her left.
Regan Graver nodded a chance just wide and McCrea had a deflected effort tipped round the post late on.
McKenzie made it four with a stunning volley on 87 minutes, acrobatically turning home her second of the day with the ball at an awkward height.
Kirsty Matthews then wrapped up an impressive performance from the Bees with an instinctive finish inside the box in stoppage-time.
Tuesday's win moved Williams' side six points clear at the top of the table going into an Adobe Women's FA Cup weekend where they travel to Reading Women in the third round qualifying.

Carly Williams reacted to her side's seventh win on the bounce following a 5-0 victory over Sport London E Benfica.
"I think in terms of our performance I was really pleased," said Williams.
"We could have been more clinical but we got there in the end. That was our biggest win of the season so I'm glad we got it over the line.
"The goal came when we started to combine. That's what we've spoken about. It was another dominant performance."
