Grêmio said centre-back Luís Eduardo has sustained a grade two injury to his right hamstring, with an estimated recovery of three to four weeks.
He was substituted after 24 minutes of the first half against City Torque on Tuesday.
His emergence followed injuries to Balbuena and Gustavo Martins. All three play on the right of the defence, creating a selection problem for coach Luís Castro.
He will miss Saturday's match against Corinthians and Brazil under-20 friendlies on six and nine June. He had been called up alongside midfield player Gabriel Mec and full-back Pedro Gabriel, both from Grêmio.
It was his fourth consecutive start for the club. At 18, he has six first-team appearances.
Pavon also came off in the first half of the draw with City Torque. Grêmio have not yet said whether the right-back has a confirmed injury.
