It was a slow start from the Bees, as Luton won an early free-kick and delivered a dangerous ball into the box. In the resulting scramble, Finley Evans reacted quickest to poke the ball past Valdimarsson.
On eight minutes, Saunders' side should have levelled the scores as a lovely ball from Maghoma found Ollie Shield, who did well to guide the ball into his path but the eventual shot just missed the target.
Shield had another effort soon after, shooting powerfully from a difficult angle but the full-back's attempt was claimed well by Henry Blackledge.
The pressure on the home side's goal grew and the leveller was found when Maghoma fired a left-footed shot from the edge of the box which fizzed past the Luton goalkeeper into the net.
Andre Grey had a great chance to put his side ahead as a pinpoint cross from Shield found his head but the flicked attempt towards goal just went wide of the right-hand post.
Arthur produced an outstanding goal-saving block to deny Dylan Stitt, who had rounded Valdimarsson but was thwarted by the centre-back’s perfect positioning.
On the stroke of the half-time whistle, the Bees put together a fine team move which saw Nunes find Carvalho in the box unmarked and the latter tapped home with ease.
The second period saw the west Londoners begin in blistering fashion as in the space of two minutes, two penalties were won and dispatched comfortably.
Donovan's skill saw him fouled in the box as the winger stood up to take the first penalty, sending Blackledge the wrong way.
Grey's strong run then saw him get tripped inside the area and this time, Peart-Harris stepped up to extend the Bees' lead as he scored the spot-kick.
The emphatic first 20 minutes of the second half was emphasised with a fifth goal, the best of the night as Konak sent a fantastic overhead kick from just inside the box into the top corner of the net.
On 67 minutes, Evans tried to add to his first-half goal as he almost wrong-footed Valdimarsson but the goalkeeper reacted quickly to adjust and save well.
Carvalho got his second of the night as he was played through on goal and struck a firm shot into the back of the net.
Arthur had a great chance to net a seventh but his free header in the six-yard box went just over the bar.
Brentford saw out the rest of the game comfortably.
Following the 1-0 defeat to Exeter City in their first PL Cup group-stage game, Tuesday’s result means Brentford B are up and running in Group E, moving onto three points.
Saunders' side are next in action on Tuesday 28 October when they face Bristol City away from home in the Professional Development League.
