Jesse Lingard scored in the first leg in Florianópolis to give Corinthians the edge. Yuri Alberto netted in São Paulo in the return to complete the job.
Internacional eliminated Athletic of Minas Gerais, winning 2-1 away, then confirming progress with a 3-2 victory in Porto Alegre.
The clubs have met in three Copa do Brasil knockouts. Internacional prevailed in 1992 and 2017, while Corinthians lifted the trophy over them in 2009 at Beira-Rio.
Reaching the last 16 has secured R$ five million in prize money, R$ two million for the fifth phase and R$ three million for this stage. If they advance to the quarter-finals, Corinthians will add R$ four million.
